Co-culture Myelination Assay

There are currently two co-culture myelination assays under development to evaluate myelination in cultured conditions. The first is a three-dimentional nanofiber culture system. In this assay, oligodendrocytes produce PLP+/MBP+ sheaths wrapping around nanofibers. The length and thickness of the wrapping correlate with the extent of myelination. The second is a co-cultured system using dissociated spinal cord cells. In this assay, oligodendrocytes sheaths wrap around axons to form compact myelin as myelination proceeds. These assays can be used to evaluate therapeutics that promote formation of compact myelin, or to screen for molecules with the potential to promote myelination from compound libraries.
