Standard and Customized Histology
Renovo offers a complete range of standard and customized neurohistology services. Our specialized histology and immunohistochemistry capabilities combined with our custom analysis techniques provide increased throughput and decreased variability for high quality tissue evaluation for central and peripheral nervous system samples. The expert scientists and technicians at Renovo have developed protocols and proprietary techniques to accommodate custom project requests for mice, rat, human or other non-human primate samples. We offer neurohistology either as a complete service or specific expertise to complement your in-house capabilities, including:
Tissue Processing: Animal perfusion, tissue fixation, and embedding (epon, paraffin, frozen or plastic).
Tissue Sectioning: Freezing microtome sections (10-100μm), cryostat sections (5-20μm), microtome sections (5-20µm), ultramicrotome semi-thin sections (up to 1μm), free floating and slide mounted sections.
Standard & Specialized Staining: Standardized protocols for traditional stains (Nissl, H&E, Tol Blue, Silver Staining, etc.) and specialized staining to assess unique features in CNS and PNS tissue.
Immunohistochemistry: Optimized protocols for several antibodies (PLP, Iba1, GFAP, MAP2, etc) as well as expertise to develop and optimize custom IHC protocols based on client requests.
Imaging: State-of-the-art bright field, epifluorescence and apotome (5x, 10x, 20x, 40x, oil 63x) imaging, automated digital full slide imaging (10x-40x), and confocal imaging.
Electron Microscopy: Automated 3-dimensional EM capabilities to interpret critical ultrastructural changes at cellular and tissue levels.
Analysis: Qualitative (morphology, overlays, reconstructions, etc) and quantitative (density, absolute counts, stereology, etc) analysis using direct, semi-automated and automated techniques.
Ultrastructural Morphometrics: Quantify and compare morphological features of cellular and sub-cellular structures including mitochondria, synapses, endoplasmic reticulum, etc.
Experimental Design & Reporting: Experts in experimental design, planning, and execution. Project reports customized to client requirements.
Neuroinflammation: Inflammation in the brain is a common feature of many neurodegenerative diseases, including PD. It is manifested by proliferation and accumulation of microglia and/or astroglia in the affected area and can be quantified by the immunohistochemical staining of microglia and astroglia markers (most commonly Iba-1 and GFAP, respectively)
Case Study: Stereological assessment of total number of Iba-1-positive microglia in midbrain
In this study, the client’s goal was to compare the neuroinflammation levels between the sham and toxin injected rats in midbrain region. We used the unbiased design-based optical fractionator stereology method to estimate the total number of microglia (visualized with Iba-1 staining) in this region.
